The BLOODLINE ; to the earth, to the moon, the clan part ways.

The O’tsutsuki clan visited the earth in quest of ShinJu fruits. Kaguya ate the fruit and subjugated the turbulent world in a blink.
After Kaguya departed leaving her 2 children behind, the younger son Hamura moved to the moon leading the (O’tsutsuki) clan and the elder son Hagoromo remained on the earth to spread 'nin shu(Nin religion)'.

BYAKUGAN; the kekkeigenkai running only in the Hyuga clan.

And later, after skipping many generations, the Byakugan Kaguya possessed was inherited (=genetic throwback) exclusively by the Hyuga clan. Since then, the Hyuga clan based on the head family, giving the top priority to the protection of this 'kekkeigenkai' have prospered.

TONERI O'TSUTSUKI
The last survivor of the O'tsutsuki clan, bent on carrying out the millenary covenant.
In order to guard the sealed Gedo Mazo, the O'tsutsuki clan have resided on the moon for a thousand long years. Toneri O'tsutuki is the last surviving descendant of the clan.
Nurtured by darkness and isolation, he sets the plan to drop the moon onto the earth into motion so as to meet the ancestral commitment that put the clan into a thousand years of darkness.
He goes into action with intentions of embarking on a life with his bride in the world after human extinction.

O'TSUTSUKI THOUSAND YEAR OLD TRADITION
The origin of all Shinobis, the O'tsutuki clan, migrated from the earth to the moon. What is the convent they've kept up over one 1000 long years...!!

THE SEAL; Sealing 10b, the moon is born
With the "Rikudoh Chibakutensei" by the O'tsutsuki brothers, Hagoromo and Hamura, the wreck of 10b―Gedomazo was sealed as the nucleus of the moon. Afterwards the O'tsutuki clan decide on moving to the moon to prevent 10b's resurgence.

THE COVENANT; A gamble with Hagoromo wishing for peace on earth
While the clan resolved on relocating to the moon, Hagoromo alone remains on Earth. Hagoromo's intention was to initiate people correctly into chakra that is essentially "power to link"
On the departure for the moon, Hamura who was assigned with the place of the head of O'tsutsukis by Hagoromo states this. "I(/we)'ll give it a thousand years." ―since then, the covenant with the big brother is strictly kept observed.

The THOUSAND YEARS DARKNESS; light shall be sealed off from the entire posterity for a thousand years hence.
Hamura was firm in his resolve. He sealed all the eyeballs of (/for) a thousand years of his (mostly unborn) descendants into "Tensei-gan" to store up gargantuan volume of power.
That was the force to be unleashed after ascertaining whether or not the earthlings in a thousand years' time are using chakra properly.
